1/3. Immunohistochemical demonstration of viral antigens in Japanese encephalitis.

    Japanese encephalitis virus antigens were immunohistochemically demonstrated in formalin-fixed paraffin sections from an autopsied brain. Glial nodules were always associated with antigen-positive cell debris. Glia shrubs in the cerebellar cortex appeared to be formed along the apical dendrite of purkinje cells. Most, but not all, of the neurons involved in neuronophagia were viral antigen positive. Antigenic masses were occasionally encountered in the center of so-called acellular plaques. neurons with strong viral antigens were sporadically found in normal-appearing regions in the thalamus and cerebral cortex. Viral antigens were demonstrable only in neurons and not in glial or vascular endothelial cells.

2/3. Serological studies on a case of laboratory dengue infection.

    One of the authors (Y.O.), who had previously been immunized with Japanese encephalitis (JE) vaccine, showed symptoms of typical dengue fever 6 days after accidental infection with a newly isolated dengue type 4 virus strain from a patient with dengue hemorrhagic fever (DHF) in thailand. His sera were examined by hemagglutination inhibition (HI), complement fixation (CF) and neutralization (N) tests. The JE N antibody titers of his sera were high even on the first day of the illness and remained almost constant during the next year. antibodies that reacted with dengue viruses were detected from a very early stage of the illness by all three serological tests. In addition, his convalescent phase sera showed high titers against all 4 types of dengue virus. These data suggest that the dengue infection caused secondary stimulation of antigens of flavivirus. Sedimentation analysis of antibodies in Y.O.'s serum (day 9) was carried out and IgM antibody that reacted only with dengue type 4 virus and homologous infecting virus was separated. These findings clearly demonstrated that the laboratory infection of Y.O. was primary dengue infection with dengue type 4 virus.

3/3. epidemiology of tick-borne encephalitis in japan.

    A tick-borne encephalitis (TBE) patient has not been reported for many years in japan although a serological survey of domestic animal sera suggested the presence of TBE foci in Hokkaido, the northern island of japan. In October 1993 in Hokkaido, a severe encephalitis case in a farm wife was diagnosed as tick-borne encephalitis. Serological examination of paired sera showed a rise of neutralization (NT) antibody titer to Russian spring summer encephalitis virus. A sero-epizootiological survey of dogs showed that the TBE-related virus was prevalent in the area. Three virus isolates were obtained from the blood of sentinel dogs and the antigenic analysis grouped the isolates into TBE-related viruses. sequence analysis of envelope protein gene identified one of the isolates as the same subtype as Russian spring summer encephalitis (Far-Eastern TBE) virus. The results provide the evidence that TBE is endemic in a certain area of japan.
